Poly (ADP-ribose) polymerase (PARP) inhibition is now the standard of care for metastatic breast cancer harboring a germline BRCA1 or BRCA2 mutation, and studies continue to investigate PARP inhibitors as monotherapies or in rational combinations. Most recently, exciting results have revealed a potential role for PARP inhibitor therapy in earlier disease settings where there is a possibility for cure, heralding a new era of targeted therapy for patients with breast cancer.
